The chemokine receptor CC chemokine receptor 5 (CCR5) has been implicated in the pathogenesis of chronic graft-versus-host disease (cGVHD). Allogeneic hematopoietic stem cell transplantation (allo-HSCT) is a curative therapy for hematological malignancies. The purpose of this study was to determine whether polymorphism in the CCR5 gene was associated with the development of cGVHD in allo-HSCT recipients. The CCR5 gene was screened in 43 patients with allo-HSCT and 62 healthy controls by direct sequencing of genomic DNA. No mutation was detected in the exon 2, 3, 4, 5 and the 3'-flanking region. The CCR5 genotype frequencies of 3' untranslated region (3'-UTR) were significantly different between patients and controls (P = 0.002). The CCR5 Delta 32 allele was significantly less common in patients than controls (P = 0.018). The three genotypes of CCR5 (Delta 32/Delta 32, Delta 32/other, and other/other) in patients were also significantly different from those in controls (P = 0.003, P = 0.001, and P = 0.016, respectively).
